That's the basic recipe for skooma, but there are other ingredients that can be added. After a few days, the nightshade will lose its essence, and simply become poison. Whatever part of the plant is used, it must be ground into a very fine powder before mixing it with the Moon Sugar, and it must be fresh. The berries and flowers of the nightshade are a middle ground, fairly potent and fairly easy to make, and the leaves of the nightshade are extremely easy to make skooma with, but the result will be very low quality, hence the name "Leaf Skooma". The root of the nightshade provides the most potent effects, but is the hardest to work with. Skooma is made from fermented sucrose (aka Moon Sugar or ethanol IRL) and nightshade.
